Abstract

Objectives:To evaluate the impact of Doxycycline (doxycycline hyclate) on the growth and viability of rabbit osteoclast-like (OCL) cell lines using a freeze thaw cycle (CT) method. Methods: The cell lines were derived from rabbit osteoclast-like (OA) cell lines that were stably transfected with a plasmid pNDC-3G-doxycycline. Results: The growth of osteoclasts, bone marrow stromal cells, and a high number of osteoclast-like cells in the CT were not reduced by Doxycycline at the early and late phases of the CT. In contrast, the Doxycycline did not affect the growth of these cell lines. Conclusions: Doxycycline treatment at early and late phases of the CT can enhance the cell cycle and growth in osteoclasts. It may be used for the purpose of inducing differentiation in osteoclasts.

Introduction

The primary use of antibiotics for the treatment of various bacterial infections has been the development of new antimicrobial agents with anti-androgenic, anti-inflammatory, and anti-oxidative effects. A variety of antibiotics have been widely used in the treatment of bacterial infections in animal models. However, the effectiveness of antibiotic therapy for treating osteomyelitis, an inflammatory condition, has not been well-established in humans. This study was to evaluate the impact of Doxycycline on the growth of osteoclast-like (OCL) cell lines. Methods: Three osteoclast-like (OA) cell lines were selected from the OA cell lines (a) in which the Doxycycline was effective at preventing the growth of OA cells and (b) in which the Doxycycline was effective at preventing the growth of osteoclasts.

Osteomyelitis is a condition that can occur when osteoclast cells are overused or overgrown. In OA, the cells become damaged by a condition called osteopenia. The loss of cells can result in an overgrowth of the osteoclasts. Doxycycline is a widely-used antibiotic and is the first-line treatment of OA. Osteoclasts are the main target of Doxycycline treatment. It inhibits the synthesis of type II collagen from type I collagen by the OA cells. However, the inhibition of type II collagen synthesis causes an increase in the level of cell surface molecules that can contribute to osteopenia. Osteopenia is caused by a variety of different factors. It is usually caused by the increased synthesis of collagen by osteoclasts and the increased production of bone resorption. Osteopenia causes an increase in the number of osteoclasts. The increased number of osteoclasts can inhibit the growth of osteoclasts. In fact, the number of osteoclasts is increased in OA patients who have been treated with Doxycycline. The increase in the number of osteoclasts may be attributed to the decreased synthesis of collagen and decreased formation of bone resorption.

The effect of Doxycycline on the growth of osteoclasts was investigated using a freeze thaw cycle (CT) method. The three osteoclast-like (OA) cell lines used in this study were derived from rabbit osteoclast-like (OA) cell lines that were stably transfected with a plasmid pNDC-3G-doxycycline. The growth of osteoclasts and the number of osteoclasts were monitored in the CT at the early and late phases of the CT and were analyzed at the conclusion of the CT. Results: In the growth of osteoclasts, the Doxycycline did not affect the growth of osteoclasts. In the osteoclasts, the Doxycycline did not affect the growth of osteoclasts. In the CT, the Doxycycline did not affect the growth of osteoclasts. In addition, there were no significant differences in the number of osteoclasts between the groups. Conclusions: Doxycycline treatment at early and late phases of the CT can enhance the cell cycle and growth of osteoclasts in osteoclasts.

Doxycycline is an inexpensive antibiotic. Doxycycline has been used for the treatment of infections and is used for the treatment of osteomyelitis. It has been found that the drug causes a decrease in the number of osteoclasts in OA.
